چکیده

The parapoxvirus orf virus (ORFV) encodes several immunomodulatory proteins (IMPs) that modulate host innate and pro-inflammatory responses to infection. Using the ORFV IA82 strain as parental virus, recombinant viruses with individual deletions in genes encoding IMPs chemokine binding protein (CBP; ORFV112), inhibitor of granulocyte-monocyte colony-stimulating factor IL-2 (GIF, ORFV117) interleukin 10 homologue (vIL-10; ORFV127) were generated characterized vitro vivo. replication properties gene deletion cell culture was not affected comparing virus. To investigate effect infection pathogenesis, groups four lambs inoculated each monitored thereafter. Lambs either or developed characteristic lesions contagious ecthyma. onset, nature severity oral commissure similar all from onset (3 days post-inoculation [pi]) peak clinical (days 11–13 pi). Nonetheless, pi onwards, regressed faster than produced by Similarly, amount shed equivalent among up day 15 pi, yet they significantly higher group 16–21 pi. In conclusion, these IMP genome resulted slight reduction virulence vivo, evidenced a duration disease shedding.

Genomes of the parapoxviruses ORF virus and bovine papular stomatitis virus.

Bovine papular stomatitis virus (BPSV) and orf virus (ORFV), members of the genus Parapoxvirus of the Poxviridae, are etiologic agents of worldwide diseases affecting cattle and small ruminants, respectively. Here we report the genomic sequences and comparative analysis of BPSV strain BV-AR02 and ORFV strains OV-SA00, isolated from a goat, and OV-IA82, isolated from a sheep. Parapoxvirus (PPV) ...

Phylogenetic analysis of three orf virus strains isolated from different districts in Shandong Province, East China

Orf virus (ORFV) is the causative agent of contagious ecthyma, which is a zoonotic disease that affects sheep, goats, wild small ruminants and humans. Shandong Province in East China is one of the main producing areas in China for sheep and goats. Here, we conducted epidemiological surveys in different areas in this Province, isolated three orf virus strains, SDLC, SDTA and SDJN, from goat floc...

Deletion of the Chemokine Binding Protein Gene from the Parapoxvirus Orf Virus Reduces Virulence and Pathogenesis in Sheep

Orf virus (ORFV) is the type species of the Parapoxvirus genus of the family Poxviridae and infects sheep and goats, often around the mouth, resulting in acute pustular skin lesions. ORFV encodes several secreted immunomodulators including a broad-spectrum chemokine binding protein (CBP). Chemokines are a large family of secreted chemotactic proteins that activate and regulate inflammation indu...

Orf virus ORFV121 encodes a novel inhibitor of NF-kappaB that contributes to virus virulence.

Orf virus (ORFV), the type member of the genus Parapoxvirus of the Poxviridae, has evolved novel strategies (proteins and/or mechanisms of action) to modulate host cell responses regulated by the nuclear factor-κB (NF-κB) signaling pathway. Here, we present data indicating that ORFV ORFV121, a gene unique to parapoxviruses, encodes a novel viral NF-κB inhibitor that binds to and inhibits the ph...

Genome analysis of orf virus isolates from goats in the Fujian Province of southern China

Orf virus (ORFV), a species of the genus Parapoxvirus of the family Poxviridae, causes non-systemic, highly contagious, and eruptive disease in sheep, goat, and other wild and domestic ruminants. Our previous work shows orf to be ubiquitous in the Fujian Province of China, a region where there is considerable heterogeneity among ORFVs. In this study, we sequenced full genomes of four Fujian goa...
